Scientists for the first time successfully replaced the "humanized" heart of a pig - RIA Novosti, 05.12.2018

MOSCOW, 5 Dec – RIA Novosti. Biologists from Europe and the USA declared a successful completion of an experiment on heart transplantation of a transgene pig in a body of several baboons. Unfortunately, donor organs are not enough for all and therefore we discuss a possibility of a ksenotransplantation long ago. About 15 years ago biologists began to discuss actively a possibility of a so-called ksenotransplantation – organ transplantation of animals in a body of the person. For the embodiment of this idea in life as it seemed to scientists earlier, it was necessary to perform simple operation – to force the immune system not to tear away "foreign" bodies. It turned out that it is extremely complex problem which cannot almost be solved even by means of gene therapy. Only a year ago the famous American geneticist George Cherch approached the solution of this task, having removed a part of such tags "-others" by means of the genomic editor of CRISPR/Cas9 and having grown up the first partially humanized pigs. © Illustration of RIA Novosti. Alina Polyanina Biologists for the first time grew up pigs with "humanized" Jan-Michael Abicht from the Munich university (Germany) and his colleagues took the following step in development of technologies of a ksenotransplantation – they successfully replaced the similar "humanized" heart in a body of a baboon, having replaced original body of a monkey. For carrying out these experiences scientists grew up own "breed" of transgene pigs at whom A3GALT2 gene which is responsible for formation of similar tags, and two other sites of DNA, CD46 and CD141 was removed, were replaced with their human analogs. Their first attempts ended catastrophically. Heart actually "died" even before change and could not support life of five baboons participating in this series of experiences. Only one monkey endured operation and lived about 30 days. This problem as scientists found out later, can be avoided if not just to keep body in a package of ice water, and to periodically pump over through it warmer blood. The following parties of baboons stretched slightly longer, about 30-40 days, however all of them equally quickly died of a hypertrophy of heart, heart attacks and other big problems with its work.
